What a happy Friday! Orbi has finished Gaozhong’s samples and is running John’s gel bands. After a nice weekend-long wash, Orbi will be running 15 various IP samples (for Scott, Kenny, and Joana).

Thanks to the OPP electricians, we have our power restored, and the instruments are pumping down. Orbi will take some time to reach operating conditions, as usual. MALDI is ready for your samples. LCT and QTRAP should be ready tomorrow.

Meanwhile, Scott’s, Joana’s, and John’s gels are incubating with trypsin overnight.

We are expecting a short interruption in our services on Wednesday, January 28 which will affect LCT and QTRAP customers (James). A longer interruption is expected for Orbitrap services (Tania): Wednesday through Friday, if all goes well. The OPP electricians will be installing a new circuit breaker in the main panel in our lab.

The sample preparation for MS (in-gel digestion) and MALDI TOF analyses will not be affected.

Orbi is running Olga’s second sample, and Ninad’s two samples are next in the queue. Gaozhong’s, Ming-Yang’s, and Yang’s samples have been analyzed and the data will be processed on Monday.

I have analyzed Alex’s oligos using MALDI TOF, next will be polymers for Brooks on Monday.

Orbi was out of calibration again, so I had to recalibrate everything; it looks much better now. All gel bands have been extracted and are running now. Tomorrow, will be the negative mode day: Alex’s oligos (provided I can calibrate the Orbi).

MALDI is bored.

The instrument was not performing to the specs during the past 2 weeks, but it looks like the issue has been resolved with the help of two Thermo tech support specialists, Mike Henry and Mohit Jain. I will check the calibrations next week to make sure everything is to the specs.

Meanwhile, Orbi is analyzing Yuning’s digest. Ninad’s samples are next. I have cleaned all gel bands submitted to date, and they are currently incubating with trypsin.

MALDI-TOF is working. I ran polymer samples for Brooks. Next are oligonucleotides for Alex and milk samples for Asha.

It’s nice to be back! Orbi is pumping down after a 2-week break. I will calibrate it as soon as the temperature stabilizes – and we will be back online! There are a few samples in the queue, but they can be run fairly quickly.

MALDI TOF survived its first holiday break in Althouse and will be running a Biotyper plate and a few different samples (oligonucleotides, proteins, gel bands) today and tomorrow.
